专家简介:Tim C. Lei博士,美国科罗拉多大学丹佛(University of Colorado Denver)分校工程和应用科学学院电子工程学副教授,生理和生物物理学、眼科学和生物工程学兼职教授;澳门大学科技学院兼职教授。本科和硕士毕业于台湾交通大学,博士毕业于University of Michigan Ann Arbor。于科罗拉多大学波博尔德分校(University of Colorado at Boulder)物理和化学系从事博士后研究工作。长期从事光物理学、超快速和非线性光学、超分辨显微镜技术、非标记成像和生物医学影像等领域的研究工作。
报告摘要:Increase of intraocular pressure inside the eye is a major risk factor for developing open angle glaucoma in which the elevated fluid pressure stresses the retinal ganglion cells leading to their death and ultimately hampering vision. The aqueous outflow system (AOS) of the eye, which consists of the Schlemm’s canal, the collector channels and the episcleral veins, is the major fluid exit pathway by which excess intraocular fluid produced at the ciliary body releases from the inside of the eye to the episcleral veins to avoid abnormal pressure buildup. Currently imaging techniques such as optical coherence tomography or confocal scanning laser ophthalmoscopy cannot image these fine tissue structures, both structurally and functionally. Therefore, it is important to develop new imaging modalities with which changes of the AOS and the retina can be imagedin order to study glaucomatous development. Multiphoton microscopy, including two-photon autofluorescence (TPAF), coherent anti-Stokes Raman scattering (CARS), second harmonic generation (SHG) and third harmonic generation (THG), uses nonlinear optical responses of biological tissuesas image contrasts to image biological samples with molecular specificity when the sample is excited by femtosecond optical pulses without the needs of exogenous fluorophores. In this talk, multiphoton microscopy techniques will be introduced and discussed, and our recent results of imaging the AOS and the retina of the eye using multiphoton microscopy techniques will also be presented.
